Le Livre D'Eli a été réalisé par Albert Hughes et Allen Hughes (From Hell, American Pimp) avec Denzel Washington (L'Attaque Du Métro 123, American Gangster), Mila Kunis (Max Payne, Sans Sarah Rien Ne Va !) et Gary Oldman (Le Drôle De Noël De Scrooge, Unborn).
